ABSTRACT

The 71Ga(n,γ)72Ga reaction-based epithermal neutron flux detectors are novel instruments developed to measure the epithermal neutron flux of boron neutron capture therapy (BNCT) treatment beams. In this study, a spherical epithermal neutron flux detector using 71Ga(n,γ)72Ga reaction was prototyped. The performance of the detector was experimentally evaluated at an accelerator-based BNCT (AB-BNCT) device developed by Lanzhou University, China. Based on the experimental results and related analysis, we demonstrated that the detector is a reliable tool for the quality assurance of BNCT treatment beams.

ABSTRACT

Objective: To explore the relationship between outdoor lighting and sports and the development of myopia, and to analyze the effects of outdoor lighting and exercise on the diopter of children with normal vision and myopia, so as to provide guidance for the prevention and treatment of myopia in children and adolescents in the future. Methods: A total of 201 children were divided into two groups according to myopia or not. Each group was randomly divided into 4 groups: outdoor exercise group, outdoor control group, indoor exercise group and indoor control group. Among them, the outdoor exercise group and indoor exercise group received moderate and high intensity aerobic exercise 3 times a week for 60 min each time for 12 months, while the outdoor control group and indoor control group had normal study and life during the corresponding period of time. No additional exercise intervention. At the end of the experiment, the diopter of each group was compared. Results: The diopter of all groups with normal vision and myopia decreased significantly after the experiment (p < 0.01). There were significant differences in diopter between outdoor exercise group and indoor control group (p < 0.01), between outdoor exercise group and indoor control group (p < 0.05), and between indoor exercise group and indoor control group (p < 0.01). There were significant differences in diopter between indoor exercise group and indoor control group (p < 0.01). The differences among myopic children after the experiment showed that there was significant difference in diopter between outdoor exercise group and indoor exercise group (p < 0.05), between outdoor exercise group and indoor control group (p < 0.01), and between outdoor control group and indoor control group (p < 0.05). There were significant differences in the changes of diopter between the outdoor control group and the indoor exercise group with normal vision and myopia before and after the experiment (p < 0.05). Conclusion: Outdoor light and exercise intervention can have a beneficial effect on children's vision, but because of whether children are myopic or not, the effect is different, outdoor light and exercise have a better effect on reducing the diopter of children with normal vision.

ABSTRACT

This study reports on the field testing of a newly and originally designed laser absorption spectroscopy chamber (LASC) system based on closed dynamic chamber method, which is well suited for multi-point synchronous measurement of ammonia emissions in field multiple plot experiment. Main design feature of the LASC system is individual multi-reflection cells for each chamber, achieving the synchronous in-situ monitoring ammonia emissions of all the chambers. Two movable covers for automated opening and closing of the chamber, and the highly transparent chamber walls made of acrylic plate minimize the disturbance of the chamber deployment on the ammonia transport process in the chamber. Controlled field assessment experiment was conducted to evaluate the applicability and reliability of the LASC system. The results indicated that the optimum time length of chamber closure for monitoring ammonia emission is 3 min, and the appropriate time length of chamber ventilation is 17 to 37 min. The LASC system has higher accuracy for measuring ammonia emission rate and reliability for comparatively measuring ammonia emissions from different treatments than the traditional chamber methods.

ABSTRACT

Understanding the fate of exogenous fertilizer-derived inorganic phosphorus (Pi) is essential for effective P management. Hence, this study carried out a 180-day incubation experiment with or without KH2P18O4 in soils with four different fertilization regimes [without fertilizer (CK), mineral P and K fertilizer (PK), mineral N, P, and K fertilizer (NPK), compost (OM)]. We analyzed the atom % excess in phosphate oxygen isotope of sequentially extracted Pi pools (H2O-Pi, NaHCO3-Pi, NaOH-Pi, and HCl-Pi), soil respiration, potential phosphatase activities, and microbial biomass. Our results showed that exogenous phosphate fertilizer was immediately transformed into the H2O-Pi and NaHCO3-Pi pools and gradually partially immobilized in the HCl-Pi pool. Additionally, biotransformation plays an important role in the turnover of fertilizer-derived Pi. After the 180-day incubation, the biologically transformed H2O-Pi content was significantly (P<0.05) reduced by 63.2 % on average, with the largest reduction in PK. The NaHCO3-Pi gradually increased in both CK and OM through biotic processes. However, it continuously decreased in PK and NPK, likely due to the strong adsorption and microbial fixation. The NaOH-Pi fluctuated slightly in CK, NPK, and OM while gradually decreasing in PK. At the end of the incubation, 28.6 %, 37.0 %, 61.2 %, and 75.2 % of the Pi increment in CK, OM, NPK, and PK were stored in the HCl-Pi pool, respectively. Overall, these findings provide important information on the dynamics of fertilizer-derived Pi, delivering new insights into rational phosphate fertilizer management and sustainable agricultural development.

ABSTRACT

Ammonia emission is one of the dominant pathways of nitrogen fertilizer loss from rice fields in China. It is difficult to measure ammonia emissions by high-frequency sampling with the chamber methods widely used in China, which is of great significance for investigating the environmental effects on the ammonia emissions. The chamber methods also can not accurately determine the ammonia emissions. In this study, the backward Lagrangian stochastic dispersion model, with ammonia concentrations continuously measured by the open-path tunable diode laser absorption spectroscopy technique, was used to determine ammonia emissions from a rice field after fertilizer application at excessive (270 kg N ha-1) and appropriate (210 kg N ha-1) rates in the Taihu Lake Region of China. High temporal resolution measurements of ammonia emissions revealed that high intraday fluctuations of ammonia emissions were significantly affected by the meteorological conditions. Multiple regression analysis showed a dominant solar radiation dependence of intraday ammonia emission cycles, especially during the rice panicle formation stage. The NH4+-N concentrations of the surface water of the rice field were found to be the decisive factor that influenced interday dynamics of ammonia emissions. Accurate quantifications of ammonia emissions indicated that the total ammonia losses under appropriate nitrogen application rate were 27.4 kg N ha-1 during the rice tillering stage and 11.2 kg N ha-1 during the panicle formation stage, which were 29.4% and 17.0% less than those under traditional excessive nitrogen application rate used by the local farmers, respectively. The ammonia loss proportions during the rice panicle formation stage were significantly lower than those of the tillering stage, which might be due to different nitrogen application rates and environmental effects during the two stages. This study indicated that the open-path tunable diode laser absorption spectroscopy technique could facilitate the investigation of high temporal resolution dynamic of ammonia emissions from farmland and the environmental influence on the ammonia emissions.

ABSTRACT

We report a facile and effective hot-pressing strategy for fabricating the flexible top electrode for a piezoelectric nanogenerator (PENG). Flexible stainless steel (SUS) foil was employed as the bottom electrode and substrate of the device. Zinc oxide nanowires (ZnO NWs) were grown on SUS substrate through the hydrothermal synthesis method. The top electrode of Zn foil was combined with dielectric polydimethylsiloxane (PDMS) film using the hot-pressing process. The resulting top electrode is thick enough to enable the device to generate piezoelectric output differently under bending conditions. The PENG devices generated an output voltage of about 2.2 V and an output current of 8 nA under the optimum operating conditions. The devices fabricated by the hot-pressing process were robust enough to retain their generating ability after thousands of bending and releasing cycles.

Reducing the applications of mineral phosphorus (P) fertilizers and supplementing them by organic fertilizers is becoming a necessary practice in the North China Plain due to overuse of mineral P fertilizers and improper disposal of organic wastes. Knowledge is needed about how the long-term substitution of mineral fertilizers by organic fertilizers affects soil P forms in order to understand soil P transformation and crop P uptake. In this study, we used solution 31P nuclear magnetic resonance (NMR) spectroscopy to characterize P forms in fluvo-aquic soil after 26 years of different fertilization management strategies, organic compost (OM), half compost in combination with half mineral fertilizer NPK (1/2 OM), mineral fertilizer NPK (NPK), mineral fertilizer NK (NK), and an unfertilized control (CK). Results showed that the P extraction efficiency using NaOH-EDTA varied from 13.0 to 27.7% for the soils of the treatments. 31P NMR spectra indicated that the majority of P was in the form of orthophosphate for all the treatments, which constituted 64.3-83.5% of the total extracted P. The application of P fertilizers significantly increased the concentrations of orthophosphate, monoesters and diesters regardless of the P fertilization method, although the proportions of monoesters and diesters were higher in CK. The proportions and concentrations of orthophosphate significantly decreased when all mineral fertilizers were replaced by compost. There was no significant difference in the proportions and concentrations of total organic P, corrected monoesters and diesters in NaOH-EDTA extracts of soils among NPK, 1/2OM and OM treatments. Decreasing mineral P fertilizers and partly replacing them by organic fertilizer in fluvo-aquic soil might increase soil test (Olsen) P and crop P uptake through the degradation of applied organic P forms.

ABSTRACT

The genome editors CRISPR/Cas9 (clustered regularly interspaced short palindromicrepeats/Cas9 nuclease-null) and TALENs (transcription activator-like effector nuclease) are popularly used for targeted modification of the mammalian genome. To date, few comparative studies have been carried out to investigate the differences between the use of CRISPR/Cas9 and TALENs in genome editing for goat breeding. Here, we compared CRISPR/Cas9 and TALEN technologies at multiple levels for generating a knock out (KO) of the Alpas cashmere goat myostatin (MSTN) gene, which negatively regulates the proliferation and differentiation of skeletal muscle cells. The electrotransfection efficiency observed using CRISPR/Cas9 was 8.1% more than that observed using TALEN for generating MSTN KO cells. In addition, the cutting efficiency of CRISPR/Cas9 for editing exon 1 of the MSTN gene was higher than that of TALENs. However, the off-target effects of the CRISPR/Cas9 system were also higher than those of TALENs. Further, we found that the frequency of obtaining MSTN-/- mutations by CRISPR/Cas9 was 8.5 times higher than that by TALEN. The CRISPR/Cas9-edited colonies involved longer deletions (up to 117 bp) than the TALEN-edited colonies (up to 13 bp). Remarkably, when embryos used to generate cloned goat via somatic cell nuclear transfer were compared, we found that the TALEN MSTN KO embryos easily developed to 8 cells and their cleavage rate was significantly higher than that of CRISPR/Cas9-edited embryos. Finally, we produced a MSTN KO lamb using CRISPR/Cas9, which suggested that a high level of targeted gene modification could be achieved in goat using CRISPR/Cas9. Taken together, our study indicates that although TALEN enables a variety of genome modifications and may have some advantages over CRISPR/Cas9, the latter provides a significant advantage by permitting precise and efficient gene editing. Thus, CRISPR/Cas9 has more potential to become a robust gene-engineering tool for application in the breeding of farm animals.

ABSTRACT

A photocatalytic reaction is always governed by energy band configuration of the catalyst, but its modulation is challenging. Here, the adjustment of band-edge positions of Ag3 PO4 through fluorescent carbon dots is reported for the first time. Both Ag3 PO4 and carbon dots which keep the similar sizes constitute a heterojunction. Such heterostructure not only promotes visible light absorption, photogenerated charge separation, and transfer, but it also transforms photocatalytic activity of Ag3 PO4 from photooxidation to photoreduction due to the huge changes of band-edge positions. In addition, the heterojunction structure of carbon dots and Ag3 PO4 exhibits unique temperature-responsive photocatalytic activities and higher photocatalytic stability than the pure Ag3 PO4 . According to the contrast experiments and related characterizations, the possible mechanism of photocatalytic reaction is proposed. Therefore, this work offers an alternative route for adjusting energy band positions or tuning photocatalytic performance as well as for preparing novel carbon-dot-based heterostructure.

OBJECTIVE: To establish a dermal sheath cell line, a dermal papilla cell line and a outer root sheath cell line from Cashmere goat and clarify the similarities and differences among them. RESULTS: We established a dermal sheath cell line, a dermal papilla cell line and a outer root sheath cell line from the pelage skin hair follicles of Cashmere goat. The growth rate of dermal sheath cells was intermediate between that of dermal papilla cells and outer root sheath cells. Immunofluorescence experiments and reverse transcription-polymerase chain reaction analysis showed that at both the transcriptional and translational levels, the dermal sheath cells were alpha-smooth muscle actin (α-SMA)+/cytokeratin 13+, while the dermal papilla cells were α-SMA+/cytokeratin 13- and the outer root sheath cells were α-SMA-/cytokeratin 13+. Patterns of cytokeratin 13 expression could distinguish the dermal sheath cells from the dermal papilla cells. CONCLUSIONS: These results suggest that cytokeratin 13 could serve as a novel biomarker for dermal sheath cells of Cashmere goat, and should prove useful for researchers investigating dermal stem cells or interaction of different types of cells during hair cycle.

ABSTRACT

Recent studies have reported the association between endothelial nitric oxide synthase (eNOS) gene G894T polymorphism and pregnancy-induced hypertension (PIH). However, the results have been inconsistent. We conducted a comprehensive meta-analysis to explore this association. A total of 36 case-control studies involving 4028 PIH cases and 7672 controls were ultimately included. In the overall analysis, no association was identified between eNOS gene G894T polymorphism and PIH risk in any of the genetic models. In the subgroup analysis, the results showed that T-allele carriers had a higher risk of PIH than those with the G allele in Asians (G vs. T: odds ratio (OR)=0.76, 95% confidence interval (CI)=0.63-0.91, P=0.002; GT+TT vs. GG: OR=1.32, 95% CI=1.09-1.59, P=0.004; TT vs. GT+GG: OR=1.96, 95% CI=1.26-3.06, P=0.003; TT vs. GG: OR=1.99, 95% CI=1.27-3.11, P=0.003; GT vs. GG: OR=1.23, 95% CI=1.05-1.43, P=0.009). For Latin American and African populations, the association between G894T polymorphism and susceptibility to PIH was only observed in the dominant model. However, no association was observed in Europeans and Americans. Therefore, eNOS gene G894T polymorphism was related to PIH risk, especially for Asians.

ABSTRACT

We evaluated the utility of chemokine MCP-3 and MDC/CCL22 as molecular adjuvants of DNA vaccines for botulinum neurotoxin serotype A (BoNT/A) in a Balb/c mouse model. Notably, the immunogenicity of the DNA vaccine against BoNT/A was not enhanced using a fusion of the AHc-C antigen with the MCP-3 or MDC/CCL22. Nevertheless, the potency of the DNA vaccine was significantly modulated and enhanced by co-administration of the AHc-C antigen with MCP-3 or MDC/CCL22. This strategy elicited high levels of humoral immune responses and protection against BoNT/A. The enhanced potency was further boosted by co-administration of the AHc-C antigen with both MCP-3 and MDC/CCL22 in Balb/c mice, but not by co-administration of AHc-C antigen with the MCP-3-MDC/CCL22 fusion. Co-immunization with both the MCP-3 and MDC/CCL22 constructs induced the highest levels of humoral immunity and protective potency against BoNT/A. Our results indicated that MCP-3 and MDC/CCL22 are effective molecular adjuvants of the immune responses induced by the AHc-C-expressing DNA vaccine when delivered by co-administration of the individual chemokines, but not when delivered in the form of a chemokine/antigen fusion. Thus, we describe an alternative strategy to the design and optimization of DNA vaccine constructs based on co-administration of the antigen with the chemokine rather than in the form of a chemokine/antigen fusion.

ABSTRACT

This study was conducted to investigate the paclitaxel loaded by hydrazone bonds in poly(ethylene glycol)-poly(caprolactone) micelles (mPEG-PCL-PTX) on proliferation and apoptosis of human lung cancer A549 cells and its possible mechanisms of anti-tumor activity. The cell proliferation was measured with MTT assay. Flow cytometry were used to analyze the cell cycle. The cell apoptosis was analyzed using Hoechst/P staining. The expression levels of apoptotic genes expression in the mitochondrial apoptosis pathway were detected by RT-PCR and Western blotting, respectively. The mPEG-PCL-PTX could inhibit the proliferation of A549 cells and promote the apoptosis. The Bax, caspase-3 protein expression were increased while Bcl-2 protein expression was decreased in A549 cells. Results showed that the polymer containing hydrazone bond is non-toxic in vitro, the mPEG-PCL-PTX micelles can inhibit the proliferation and induce the apoptosis of A549 cells. The backward Lagrangian stochastic dispersion model in conjunction with open-path tunable diode absorption spectroscopy was used to quantify ammonia emissions from farmland based on the high-temporal resolution data, aiming to provide innovative achievements to diagnose patterns of ammonia flux. The results indicate that the bLS dispersion technique using open-path lasers to measure atmospheric ammonia concentrations is suitable for determining ammonia emissions from farmland continuously, especially for characterizing diurnal characteristics of NH3 emissions. The ammonia emissions have a significant diurnal pattern with two emission peaks from urea applied to maize on a calcareous sandy loam fluvo-aquic soil in the North China Plain. We believe that the first peak starting at approximately 9:00 am is due to NH3 absorbed by the dew re-emission at night as the dew evaporates. The maximum of ammonia flux at 14:00 corresponds to the peak of soil temperature and solar radiation. The ammonia emission increased rapidly, but the duration of emission peaks lasted approximately 4 d. Cumulative NH3 emission was 25.3% of the applied N over the entire measurement period. The NH3 emissions measured with bLS dispersion technique and venting method had certain difference.

ETHNOPHARMACOLOGICAL RELEVANCE: Danshensu is an active water-soluble component from Salvia Miltiorrhiza, which has been demonstrated holding multiple mechanisms for the regulation of cardiovascular system. However, the relative contribution of danshensu to its multiple cardiovascular activities remains largely unknown. AIM OF THE STUDY: To develop an artificial neural network (NN) model simultaneously characterizing danshensu pharmacokinetics and multiple cardiovascular activities in acute myocardial infarction (AMI) rats. The relationship between danshensu pharmacokinetics (PK) and pharmacodynamics (PD) were evaluated using contribution values. MATERIALS AND METHODS: Danshensu was intraperitoneally injected at a single dose of 20mg/kg to AMI rats induced by coronary artery ligation. Plasma levels of danshensu, cardiac troponin T (cTnT), total homocysteine (Hcy) and reduced glutathione (GSH) were quantified. A back-propagation NN model was developed to characterize the PK and PD profiles of danshensu, in which the input variables contained time, area under plasma concentration-time curve (AUC) of danshensu and rat weights (covariate). Relative contribution of input variable to the output neurons was evaluated using neuron connection weights according to Garson's algorithm. The kinetics of contribution values was also compared and was validated using bootstrap resampling method. RESULTS: Danshensu exerted significant cTnT-lowering, Hcy- and GSH-elevating effect, and these marker profiles were well captured by the trained NN model. The calculation of relative contributions revealed that the effect of danshensu on the PD marker could be ranked as cTnT>GSH>Hcy, while the effect of AMI disease on the PD marker could be ranked in the following order: cTnT>Hcy>GSH. The activity of transsulfuration pathway was quite obvious under the AMI state. CONCLUSION: NN is a powerful tool linking PK and PD profiles of danshensu with multiple cardioprotective mechanisms, it provides a simple method for identifying and ranking relative contribution to the multiple therapeutic effects of the drug.

ABSTRACT

AIM: To investigate the pharmacokinetic interactions induced by content variation of the main water-soluble components of Danshen injection in rats. METHODS: Intravenous Danshen injection (control) or Danshen injection with danshensu (DSS), protocatechuic aldehyde (PAL), salvianolic acid A (Sal A) or salvianolic acid B (Sal B) were administered to female Sprague Dawley rats. Plasma concentrations of DSS, Sal A, PAL and its oxidative metabolite protocatechuic acid (PA) were analyzed simultaneously with LC-MS/MS; concentrations of Sal B were determined by the LC-MS method. Non-compartmental pharmacokinetic parameters were calculated and compared for identifying the pharmacokinetic interactions among these components. RESULTS: Compared with the control group, the DSS, Sal A, and Sal B groups had significant increases in AUC(0-infinity) in response to elevated concentrations of PAL (by 78.1%, 51.0%, and 82.9%, respectively), while the clearances (CL) were markedly reduced (by 42.5%, 32.9%, and 46.8%, respectively). Similarly, Sal A increased the AUC(0-infinity) of DSS and Sal B (26.7% and 82.4%, respectively) and substantially decreased their clearances (21.4% and 45.6%, respectively). In addition, the pharmacokinetics of DSS and Sal B were significantly affected by the content variation of the other major components; the AUC(0-infinity) increased by 45.1% and 52.1%, respectively, the CL dropped by 29.6% and 27.1%, respectively, and the T(1/2) was decreased by 22.0% and 19.6%, respectively. CONCLUSION: Complex, extensive pharmacokinetic interactions were observed among the major water-soluble constituents in the Danshen injection. The content variation of PAL had the most significant effect on the pharmacokinetic behaviors of other major constituents. Furthermore, the pharmacokinetics of DSS and Sal B were the most susceptible to the content change of other components.

ABSTRACT

In this paper, a new dual fluorescent N,N-dimethylaminonaphthalene derivative, sodium 4-(N,N-dimethylamino)naphthalene-1-sulfonate (SDMDNS), was reported. It was found that SDMDNS emits dual fluorescence only in highly polar solvent water but not in organic solvents such as methanol, dioxane and acetonitrile. Only a single broad band emission at ca. 420 nm was observed in the short wavelength region in organic solvents. The dual fluorescence of SDMDNS in water was found at 423 and 520 nm, respectively. Introduction of organic solvent as ethanol into aqueous solution of SDMDNS leads to blue shift of the long-wavelength emission, and this was evidently supported by introduction of cyclodextrin or surfactant in the aqueous solution. It indicates that a highly polar solvent was required to bring out dual fluorescence; furthermore, the short wavelength fluorescence is emitted from locally excited (LE) state and the long wavelength fluorescence is emitted from charge transfer (CT) state. The pH dependence of the dual fluorescence of SDMDNS demonstrates that the neutral form of the molecular has a higher ratio of CT band intensity to LE band. Temperature effect on the excited state of SDMDNS was also examined and gave stabilization enthalpy (-DeltaH ) of the CT reaction 8.7 kJ mol(-1).
